The Zeiss LSM 710 NLO microscope is based on an Axio Examiner.Z1 microscope combining confocal and multiphoton microscopy, with 2 Mai Tai eHP DeepSee “Ti:Sapphire” Infra-Red lasers, 34 spectral R/FL detection channels, Transmitted T-PMT, BIG detectors as NDD and Transmission NDD, and 6 laser lines 405, 458, 488, 514, 561 and 633nm. Axio Examiner.Z1 is an upright fixed stage microscope and the platform for the LSM710 NLO for intravital imaging and electrophysiology. It is especially suitable for Multiphoton Imaging of thick tissue and whole animals (intravital imaging). The Mai Tai® DeepSee™ “Ti:Sapphire” is a tunable laser (690-1040 nm) with over 2.4 W of average power. The Mai Tai oscillator has automated dispersion compensation that has been specifically optimized for use in multiphoton microscopy, allowing deeper penetration depths without the risk of sample damage from increased average powers. Moreover, the LMOA platform provides equipment to perform electrophysiology measurements such has: 2 motorized micromanipulators, a signal amplifier (Multiclamp 700B), signal digitizer (Digidata 1550) and software (pClamp10). This innovative system provides means to pursue very complex methods such as two-photon uncaging in connection with calcium imaging. The LMOA platform also provides infrastructures for cell culture and maintenance of small animals.
